Adult stem cells are extremely important for the robust generation of major specialized progenitor cell types and thereby remarkably maintaining the tissue homeostasis throughout the adult life. Whereas, ionizing radiation (IR) exposure to hematopoietic system could intrinsically damage the stem cell functional capacity, leading to defect in reconstitution and proliferative potential, diminished self-renewal, elevated apoptosis and, eventually, functional exhaustion of the stem cell population. It has been suggested that accumulated DNA damage could be a principal mechanism underlying stem cell decline. In adult hematopoietic stem cells (HSCs), radiation induced elevated levels of DNA damage and/or oxidative stress may have profound impact on the functional capacity of the stem cell population. Although several reports characterized the DNA damage patterns in HSCs, it remains unclear how hematopoietic stem and progenitor cells (HPCs) respond towards radiation induced DNA damage. In order to achieve this goal, we are extensively investigating the underlying DNA damage patterns that trigger genetic instability in HSC and HPCs. Our main objective is to investigate how radiation stress could potentially alter the proliferative, self-renewal, cell cycle characteristics of hematopoietic stem cells. Using a mouse hematopoietic system, we are studying the DNA damage induction and repair patterns in stem and progenitor cells after the radiation insult in vivo and in vitro. Our aim is to establish that radiation induced stem cell injury could be at least, in part, rescued by mitigating the elevated levels of DNA damage and or oxidative stress in HSCs. More importantly, our study will establish that DNA damage and or oxidative stress accrual may contribute to transformation of stem cells into cancer stem cells with increased genetic instability. Unraveling the potential mechanisms of severe DNA damage and or oxidative stress could directly explains stem cell behavior under stress and possibly provide key insights for the development of a novel mechanism-based therapeutic strategy to mitigate radiation-induced stem cell injury.
